I have reported that chemotherapy for the patient with Li-Fraumeni syndrome results in the ectopic CD44 variant-positive cancer stem cells. Given that chemotherapy often increases the ROS level, excessive oxidative stress influences on the splicing pattern of CD44, thereby increasing the synthesis of CD44 variant. Does anyone have the similar observation?
Over 20 years ago I helped to elucidate the genomic organization of the CD44 gene in the mouse system (but to my big surprise the highly questionable lab did not include me as coauthor); at that time, many labs produced then data on CD44 variants and tumor metastasis - but it seemed to me and to many people in the lab that many of the original data and interpretations were partly questionable or inconclusive - basically except the existence of the gene.
I am wondering how much is clear today about CD44 variants in any cancer.
ESRP1 (Epithelail splicing regulatory protein 1) promotes the generation of CD44 variant 8-10, which includes variable exons 8-10, which is highly expressed in cancer stem cells.
