When you isolate endothelial cells of the blood vessels, using only CD31 antibody is not enough to isolate them? Why alpha-smooth muscle actin (alpha-SMA) antibody is necessary according to some of the literatures?
What kind of cellular population is contained in CD31+/alpha-SMA+ population?
On the other hand, furthermore, what is the cellular population of which the expression pattern is CD31-/alpha-SMA+?

People use certain cell surface makers to isolate a specific cell lineage for the gene expression study or the development of therapeutics to kill cancer cells or the regeneration of damaged tissues or the demonstration of feasibility of tissue engineering. The varying biomarkers employed reflect different studies for their own aims.
Thus, your aim has to be specified as mentioned above why you are isolating a specific cell lineage expressing CD31/a-SMA. Depending on the aim, you may need CD31 alone or CD31 and other cell surfaces makers together or as you read.
In considering the origin of endothelial cells and hematopoietic cells in development of the embryo, the origin is hemangioblast stem cells. While the blood vessels get a mature and functional forms, mesenchymal cells (once epiblast, then mesenchyme, some of which becomes mesoderm and endoderm population).
Mature vessels have a fine, complex structure consisting of endothelial cells, smooth muscle cells, myofibroblasts (pericyte progenitors) and other mesenchymal cells, etc.
Thus, CD31/a-SMA dual makers are used for the verifying vessel-consisting cellular components. HSC makers, CD31 or CD45, are not enough to take all cellular components of the thinner vessels, eliminating pericyte progenitor, myofibroblasts etc. These cells, in fact, can be obtained from trans-differentiation occurring in vivo and in vitro, suggesting mesodermal origin of the blood-forming and vessel-forming cells.
