So far what I have read in Leishmania proteome for exosome I know that density gradient ultracentrifugation is the best method to get the exosome. You will need to perform western from the pellet proteins and look for the markers of exosomes. Also you can do imaging to see actual exosomes. Please follow this link and you will feel happy.

Hi Malak, Have you tried any of the exosome isolation methods? Roughly what are the yield?

Be careful using serum. You will get a higher and more pure yield than with plasma but a surprisingly large population of the exosomes in serum will have been formed during the clotting process so may skew your results.

ExoQuick by System Biosciences works very well with small samples, as does a similar binding buffer for exosomal isolation from plasma from Invitrogen, my yield has been similar using both methods in human and rat samples.  The number of exosomes isolated does vary from sample to sample, and there are likely a number of factors that contribute to that.

Invitrogen's Total exosome isolation reagent works well- there are 5 options, for misc sample types (serum, plasma, cell media, urine etc)
