Now i am working on AM fungi single strain. What are the most effective isolation techniques for obtaining pure cultures of arbuscular mycorrhizal (AM) fungi?? Is there are best approach techniques for isolation that increase chance infection or colonization AMF?? (AMF from single strain from consortia or soil)

To get single strains of AMF, one of the most appropriate ways is to establish single spore cultivation assays. Spores may originate from field soil samples or pot trap cultures based on soil, root, or root + soil samples.
It is very fun to work with AMF, such wonderful soil microorganisms with the biggest spores ever. I suggest you to read the recent books describing simple techniques for in vitro cultivation of AMF.
