Hello everyone, 

I am planning to assess the effect of a drug on cancer stem cells proliferation. Giving that the mammosphere assay is an efficient way to assess cancer stem cells proliferation, I woud like to know which method is experimetally appropriate: treating adherent cells with the drug and then using surviving cells to generate mammospheres or generate mammospheres from non treated cells and then treat mammospheres with the drug.

I would be really grateful for sharing opinions and experiences.
