Hello everyone,
I have a trouble in detecting the active form of caspase 3 and 9 using western blotting. Despite being successful in detecting the cleaved form of PARP, I failed to detect caspase 3 and 9(active form) . Any recommendations?
I transfer my proteins to a PVDF membrane for 30 min at 100V.
Loaded protein amount: 40 microgram.

you chek with different interval of treatment time like 6h, 12h, 24h and run the gel as soon as possible after lysate preparation at low temperature.
