I want to study the effect of EGF on the expression of a target protein. Is there any alternative to serum starvation? Do you think that treating cells with a high concentration of EGF in a 10 % FBS containing medium might solve the issue?

I have used the following in working with human diploid fibroblasts. I used a pretreatment of medium lacking EGF before the experimental treatment:
"Basal medium MCDB-104 is supplemented with platelet-derived growth factor (30 µg/ml), epidermal growth factor (100 ng/ml), insulin (5 µg/ml), transferrin (5 µg/ml), and dexamethasone (55 ng/ml). With this medium cells will grow at a rate and to an extent similar to that produced by medium containing 10% serum. During one growth cycle the cultures can accomplish up to seven population doublings. "

May I ask why you have added Dexamethasone (55 ng/ml) to the medium?
