I am evaluating the potential anti-invasive effect of cytotoxic compound on breast cancer cells, MDA-MB-231 and SK-BR-3. I ordered Cultrex 96 well BME invasion kit and the manufacturer recommend that the cells should be passaged 2 or 3 times prior to use. My question is do they mean we should use cell that passeged only 2-3 times after thawing the original vial or we can use cells that had been passaged several times (for example, more than 10 times) in order to confirm their viability and growth pattern?
