I have some questions related to WST-1 cell proliferation assay.
I did a WST-1 assay experiment with my pancreas cancer cell line in 96-well plate. First, I plated the cells in 96-well plate and the next day I treated cells with a drug called Luteolin. After 24H, i also treated the cell with human protein called TRAIL. 48H later, I checked the absorbance measurement with plate reader, but the result does not come out.
NB: With microscopy, we could see that the cell number decreased as the drug concentration increased. But the result from the absorbance measurement show no difference in cell number for all wells.
1) What possibly make the WST-1 error?
2) What is a good condition for WST-1 assay, like the temperature, the absorbance length, etc.
3) Is WST-1 assay working to almost cell lines?
