I have some questions related to western blot as I am new to western blot.

I tried to detect the LC3B, however I got diffused bands as I have attached in the file (Please find the attached file below).

Here is the flow of my experiment.

> I add 20g of protein per well.

> I used Q-PAGE TGN (Tris-Glycine Novel) Precast Gel (#QP4510)

> Running speed and time: 60 constant volts for about 1H50 mins

> Transferring speed and time: 120 A for about 2H00

> Membrane: Nitrocellulose membrane (NC)

> Primary anibody: mouse monoclonal LC3B (sc-271625) --> 5ug into 20mL 1% skim milk (1:4,000)

> Secondary antibody: Anti-mouse IgG HRP-linked antibody (cell signaling 7076s) --> 3ug into 15mL of 1% skim milk (1: 5,000) per membrane

Have anyone experienced this kind of result before?

I would like to know are there any possibilities that can cause these diffused to appear? And any suggestion for trouble-shooting techniques?
