I use the miR Data Intergration Portal (miRDIP) to look for predicted targets of a specific microRNA. Here's the link to the website - http://ophid.utoronto.ca/mirDIP/ It draws target prediction data from several different sources and gives you a compiled list and the database it obtained the prediction from. I use that information to decide which targets to run qPCRs for. Its a lot faster than checking individual target prediction websites.

For the primers, you could either design them, look for publications that have validated the qPCR primers or look for them in this primer bank (https://pga.mgh.harvard.edu/primerbank/)

For predicting the targets a number of online and standalone tools are available e.g.

mirBASE- TARGETScan

DIANA

miRwalk

miRdb

comiR

Please make sure you understand the criteria for sorting the results e.g. CWCS in case of targetscan.

You can initially confirm the target by checking the complementary between your miR and target mRNA and later by qRT-PCR for your transfection experiments

FYI: For designing primers a very easy to use tool miR primer is available for download.
