I accidentally left my tissue (mouse pancreas/duodenum/lung/spleen) in PFA overnight instead of 30% sucrose. Typically, I fix the tissue in 4% PFA for 45-60 minutes and then 30% sucrose O/N. I then freeze in OCT and section at a later date for IF/IHC.

Just realizing my mistake I just placed the tissue in 30% sucrose. When I section this tissue in the future will I need to take any extra steps for antigen retrieval due to the extended time in PFA?
