I have worked on invivo toxicity of liver kidney now i m shifting towards toxicity in mammalian cancer cell line. Can i use protocols i already have?
Thanks.. Even many commercial kits describe sample preparation for tissue in mg and cells in number... But in my case hw to convert mg in cell number.. Some article has suggested that 1 gm of cancer tissue pocesses 1*10 (9) cells.. But i doubt this will be standard way of conversion or not
Thanku Andrey.. sir, may i correlate it with the amount of total protien concentration in given sample? Like... In x amount of protein (sample source b either cell or tissue) y amount of glutatione / SOD/ catalase is present??? So can i corelate enzyme production with total protein????
Well...i think m not expressing my query properly.. Let me try... Like in tissue sample (treated), the activity of GSH is showed at "y" concentation of total protein... Now in cells i have to take atlest "y" amount of protein to get activity of gsh... so first i will calculate protein in cells of interest than compare it with the total protein in tissue... Then take sample volume(cells protein) for GSH assay according to the protein required.
For this i only need to standarlized or focus only on total protein or compare tissue and cells in the context of protein concentration(not the amount of tissue vs cell number).... Then follow the protocol as same as in tissue..
No no i m not expecting the amount of gsh will be same.... I am just figuring out that if amount of protien will help me to caliberate my method from tissue to cell... However i know cancer cells possess low concentration of antioxidant.... n thanks alot for your help.
