The graph show cyctoxicity of different combination of chemo drugs on a cancer cell line. The graph compare the percentage of cancer cell growth inhibition and percentage of cancer cell killing with respect to different concentration of drug.
The line parallel to x-axis (at 0% cell growth) is the line of total growth inhibition (TGI). Above TGI, cell growth is only inhibited (without any cell killing) and below TGI, in addition cells are actually killed.
Usually I do MTT Assay to find the percentage of cell viability. My question is how one can calculate percentage of cell growth inhination and cell killing simultaneously?

That is not possible using MTT !
If you want to state on the "cell death" effect of your molecule, you will have to perform some Annexin analysis, or WB of apoptotic markers.
In my lab we use proliferation assays, since our cell counter allows for the quantification of dead cells vs live cells.

As say you David is not possible using MTT calculate cell death. MTT show you the viability but you dont know whta happen with your cells at middle time, you only know about final viability.
You need to perform Annexin-IP analysis to measure cell death. In my lab we use toluidin blue dye but iti is a hard work when you has different cell lines or various treatment drugs or both.
