I am loading Gemcitabine HCl into mesoporous silica nanopariticles. I took 25 mg of mesoporous silica nanoparticles and 10 mg of drug for loading purpose. I used PBS 7.4 as a medium to load the drug. The gemcitabine Hcl drug has negative charge in water at PBS 7.4 is -7. The mesoporous silica is also having negative charge in water at PBS 7.4 around -18. I am getting drug loading percentage around 2% only. The solubility of gemcitabine in water is around 25 mg/ml. My question is that
1) solubility of drug in particular solvent playing a major role?
2) pH of the solvent playing a major role because
at pH 4,
Charge of mesoporous silica is around -15
Charge of gemcitabine is around 12
so, If I use PBS 4 as medium for drug loading, there will be more drug loading is possible?
Yes and the pH and ionic strength of the solution will be critical. Lower the pH and reduce the amount of dissolved salts if possible.
Despite this if drug release studies are to be performed on pbs buffer I can assure you a fast release.
both drug solubility and pH play important roles. optimising the pH would definitely help
even if you lower the pH for practical application in intracellular delivery when you apply your drug loaded nanomaterial into the media it will release instantly . So I suggest you to make positively charge mesoporous silica by functionalzing it with positve charge it will give you good results with binding . All the best

Making the silica positive will be good for nothing given the nature of your drug.
Making the silica positive has so many advantages,
1) More drug loading is possible
2) One can attach polymer or targeting ligand over it
3) silica will release more drug in acidic pH compared to normal physiological pH 7.4

It is just a suggestion only ... both ways can be tried changing pH as well as charge reversal, one can choose whatever gives best results for the ultimate experiments. Thank you.
