I am working with mesoporous silica nanoparticle for the treatment of cancer. I am interested to know how much amount of mesoporous silica nanoparticles internalized in cancer cells. I seed 1 million cells in T25 flask and allow the cells to attach with surface for 24 hrs. After 24 hrs, I added 200 mg/ml of mesoporous silica nanoparticles for internalization. I washed cells with PBS after 8 hrs and did centrifugation followed by trypinization. I don't know how to dissolve the mesoporous silica in cancer cells for ICP-AES analysis? Looking for good suggestions.

In my opinion, the silica could be solved in the hydrofluoric acid. It is necessary to use plastic vessel for this operation, because the hydrofluoric acid is very aggressive towards the glass.

Thanks for your answer. But in some papers, people are using 3 ml of Conc Nitric acid to cancer cells to dissolve the mesoporous silica and then heat the mixture to 50 degree celsius for 2 hrs. Finally make the solution to 15 ml using distilled water. Then submit the sample for ICP AES analysis.

it was misunderstanding, procedure described by you will not dissolve the mesoporous silica but it will extract the compounds for ICP-AES from the silica, which is probably your aim.

Dear sir,
My aim is to find how much amount of silica present within the cancer cells sir. People are using 4 M NaOH , CONC. Nitric acid, HF to find the amount of silica content within the cells. I tried with conc.nitric acid once, results were too good sir.
Note: I am doing my PhD currently. so Just prabhuraj enough sir.

I don't have a direct answer to your question but would like to know how you dissolved the Mesoporous silica nanoparticles. What solution did you use (PBS, DPBS,?) Also did you get a uniform solution or a suspension. Did the particles settle/ separate out from the solution after kept standing?
