We are doing a resistant cancer cell line through multiple exposures to a certain chemotherapeutic drug. Considering that the cell morphology had also changed, how would you know if the experimental cells were of the same cell line (as the parental) and how to determine if the current cells are not cross-contaminated with the other cell lines?
You can study Cell cycle analysis of drug treated cells and untreated cells by flow cytometry and check DNA distribution in different phases.
For sure, the morphology change during this process. You can check the related articles, working on the same cell line to see if the morphology of the resistant cells is similar. STR profiling is the best approach for characterizing the cells.
The alternative approach can be use some cell specific surface markers through ICC.
