The working volume for 24 well is 1 ml, I should standardized the medium below 1 ml. For example, I will put complete growth medium (CGM) about 500 ul in each 24 well plate. But when I added the treatment solution into each well by the specified volume, this will cause the working volume for each well changed. Can the total volume of medium in each well are different,? CGM's volume is same for all wells.
The best is to keep changes minimum in biology. Playing with volumes to keep the end volumes constant may be a good idea but it all depends on the readout involved.
If you can dissolve the treatment substance in CGM, then make various concentrations of the substance in CGM, you can vary the final concentration of the substance while keeping the total volume and concentration of CGM constant.
How about the treatment substances in concentrated liquid? This will causes the working volume in each well differ because I will put the volume of concentrated liquid varied based on desired concentration. Is it possible to add the concentrated liquid without applying the freeze-drying or lypholized techniques?
As Adam B Shapiro already suggested keep the volume constant and calculate in which concentrations you have to prepare your treatment substance to achieve the final concentrations required. If you have a highly concentrated stock of your substance and only need to add a few microliters of it that will have no impact on your experiment. If you don't know how to do the math for that ask your supervisor.
I already have math for it. Hopefully, the concentration liquid does not affect the CGM much. Thank you all for your opinions.
