Hi,
Most of the research papers or protocols mentioned that DMEM should be supplemented with 10% FBS or FCS for the cell culture. However, it does not mentioned clearly 10 % in the DMEM or else. Can someone clarify me this. I have treatment media in liquid state which will cause DMEM volume to decrease a bit. So, I am considering to prepare 10% FBS either in the DMEM or working culture medium.

Most of treatment samples usually been added to media in the form of extract or powder. In my case, the sample should not be extracted or lyophilized etc, because will degrade its quality. The total working volume of my cell culture is 1ml per well (24 well). Thus, I will put let say 0.29ml of treatment sample and 0.71 of media culture. But, I am considering either to add 0.71ml complete growth media containing DMEM, 10% FCS, and pen-strype, or the volume of 10% FCS should be added into 1 ml of total working media. Hope you understand my point.

When we make a complete medium then we consider adding 10% FBS in DMEM and other supplements in DMEM only.
Let me make it simple for you, e.g. if I am making 100ml treatment media then 10% FBS will be 2ml FBS + supplement A (2%) 200ul, supplement B (5%) 500ul, supplement c (3%) 300ul....so it will be total 2ml FBS+200ul A+ 500ul B + 300ul C=3ml+97ml DMEM= 100ml complete treatment media. That's how you make the complete treatment media. some ul of volume like 10-20ul doesn't matter much in such big volume of DMEM but to be very precise about your exxperiments, you should also take that into consideration.
