I need to measure short chain fatty acids from my experiment, but my problem is limited volume of sample I have during the experiment. I know for HPLC of short chain fatty acid 20 microlitre would be enough but as samples need to be centrifuged first I dont know how much is the minimum amount of sample that I need to take and centrifuge for 1 replicate of HPLC analysis ( ofcourse I know for 2 replicates of HPLC it will be doubled)
Depends on your sample type (plasma?), the expected concentration of SCFAs in your samples and the detection limit of your method.
If the concentrations are far above the LOD/LOQ, you may collect just a few microliter and dilute your sample to an appropriate volume.

There are a number of different methods. A good method should have limits of detection and quantification and a sample preparation procedure which should answer your question.
