i am about to seed a T75 bottle with different dilutions ( 1:2 and 1:10 ) of 100% confluent Huh7.5 cells(about 7-8X10 5 cell /ml ) ............ how long do those seeded cells take to reach 100 % confluency ? is there a specific formula to calculate the rate ?
I also worked with Huh-7.5 cells in the past. I think it depends on the medium you use for culture. I used DMEM + 10%FBS + 1%MEM + 1% L-Glutamine and my cell passage is lower than 15 times. If I split at 1:2, they will reach confluence just nearly 2 or even 1 day; at 1:5, they will reach confluence after 2 or 3 days and at 1:10 around 5 or 6 days.
I have seen these cells reaching to 100% confluency in 2 days, if split at 1: 2 ratio.
In my experience cell number increased approximately 1.5-fold per day
I always split these cells at 1:4, they would reach 100% confluency in 2 days.
