I want to do a coagulase test for the identification of staphylococcus aureus. When I asked how to get the "rabbit plasma " and what is the most appropriate procedure for doing so, someone suggested that I would get human blood and centrifuge it to separate plasma and use it as my reagent rather than purchasing the standard "rabbit plasma" , especially because I am about to survey a large number of isolates. Is this a realistic suggestion?
Yes, you can. But you need to take some considerations:
1. S. aureus strains are generally capable of coagulating citrated or EDTA-treated plasma in the tube test and will produce clumps of cells in the slide test.
2. The use of fresh, citrated or EDTA-treated human plasma is not recommended because of the risk of infection.

Yes you can use human plasma for a huge load of coagulase test, but preferably pooled and previously HlV and hepatitis tested negative.
Diluted rabbit plasma is the the accepted one for confirmation of S. aureus.
Strains of human source are usually positive with both human or rabbit plasma.
But the animal strains, S. hyicus and S. intermedius react only with rabbit plasma.
