I am try to find a way to differentiated the cancer cells to a point where they are terminally differentiated. They are similar to Neural stem cells which is why I removed all of the growth factors and only added serum + dmem/f12. Is there any other chemical that I can add to see if these cells will further differentiate.

Try adding some fibroblast growth factors (FGFs) and epidermal growth factor (EGF) to the medium. And what kind of serum do you put into the media? Fetal bovine serum (FBS), horse serum (HS), B12 etc.?

Yes, add to medium 20 ng/ml of basic fibroblast growth factor (bFGF)& 20 ng/ml of epidermal growth factor (EGF, Peprotech) .
For culterure cell : DMEM/F12, supplemented with antibiotics (penicillin 100 IU/ml, streptomycin 100 µm/ml), HEPES (10 mM), glutamine (2 mM), glucose (0.3% w/v), recombinant human epidermal growth factor (rhEGF) (20 ng/ml), basic fibroblast growth factor (bFGF, 20 ng/ml) and N-2 Plus Media Supplement (Neural Stem Cell Medium, hereafter named: NSCM). The medium was changed every 2 days.
