With the help of researchgate´s community I found that to determine if a cell line is clinically-relevant resistant there needs to be a 2 - 5 fold change increase in the IC50 values of the cell line tested when compared to the IC50 value of the parental cell line.
Could we used the same approach (considering a decrease instead of a decrease to determine if a cell line is sensible or not?
but the paper explaining that, did not specify if the same could be applied for a sensible cell line.
Could this method be applied for cancer cell lines? If it does, which would be the best option to use as a parental cell line? Our lab has the MCF-12A cell line and i was thinking of using this as my "parental" cell line.

Maybe you can consider cell viability assay with drugs in continuous concentration to determine how the drug influences cell proliferation. After data processing, an IC50 will be got by fitting the curve. If your cell line is sensible to a drug, with higher generation of your cell line, the IC50 may increase or decrease. Our group often test a drug's activity on prostate cells. Hope my answer helps.
It all depends on the range of concentrations you use to determine your IC50. All cells will usually respond to high doses of drugs. For a clinically meaningful IC50, you'd want to have an IC50 < 1um. I recommend that you research the literature for the drug in question to arrive at sensible data in terms of drug response.
I agree with Carrie: I also use breast cancer cell lines and for determining the IC50 I use different drug concentrations and I evaluate either with viability assay or with clonogenic assay. As parental cell line, it depends on which kind of breast tumor (depending on the classification based on the gene Expression profile) you want to study, especially because they are sensitive in a different way: for example the triple negative is the most aggressive, so you should always compare each cell line to the respective parental one.

It depends on the drug, route of administration and kind of assay, whether it is in vivo or in vitro, or if it is previously done in animal experimentation in order to be able to extrapolate the results into humans. In a cell culture it can be chacked for growth or inhibition. In an animal test the same can be verified histologically and if there is cellular differentiation. At the electron microscopy level RER enlargement and increase or depletion of secretory granules, or increased apoptosis, or frank necrosis can be verified. At the biochemical level it can be checked by looking for substances that release a certain type of cell. Finally, there is a range of possibilities and techniques at the experimental level to be able to determine wheter or not a tissue is sensitive to a determined drug.
