I´m new to cell culture and i was wondering if this morphology (image 1) is common when growing MCF7 cells and if clustered cells (image 2) give rise to changes in the cell line phenotype for further studies (unappropiate homogenization during detachment with tripsin).

That's something really common specially for some immortalized or cancer cell lines. If you're worried about, simply don't. Often cells slightly change their average shape and behavior by simply change the culturing surface from plastic to another brand's plastic or from plastic to glass. There are also cell lines I used where you're likely to find tremendous differences in size and shape from cell to cell in their neighborhood, heterogeneous ploidy and nuclei size etc. They also often change their response to treatments depending upon kind of media used and nutrients richness, cells density, the above mentioned growing surface and so on. Just consider all these possible variables and do your experiments
MCF-7 is a kind of cells lacking contact inhibition, so it simply has no limitation of proliferation in normal culture dishes. as a result, it changes its shape and size in the different confluences.so I guess it's normal when you observe different cell morphology. usually, i pass this cell in 70-80% confluence and divide 1/4-1/5 in passages so that the density would not be too low or high.

Domes are common in mcf7 you should avoid to reach confluence. But anyway when you will treat the culture with tripsin they will separate well with no problem for further assays. Remember yet in the 80's has been described that in mcf7 coexist at least 5 subpopulation with diferent molecular profile, so how the cell line is.
