I am working in cell culture using Tk6 cells and primary cells (human lymphocytes). Analysing the global DNA methylation with an ELISA I observed only 0.1-0.2 % of global methylation status. However, this is way too low as the DNA methylation should be between 2-5%.

Does anyone know if it is possible to loose global DNA methylation due to wrong culturing techniques (media) or during DNA isolation, which was done using a Quiagen Isolation kit.
