Has anyone observed loss of global DNA methylation in HuLy or Tk6 cells without treatment - either due to wrong culturing or during DNA extraction?

I am working in cell culture using Tk6 cells and primary cells (human lymphocytes). Analysing the global DNA methylation with an ELISA I observed only 0.1-0.2 % of global methylation status....
