For one particular construct we are trying to clone, we cannot get colonies to grow in culture. We are ligating a DNA fragment into a vector, transforming the ligation mix into XL1 Blue cells and plating out. We get colonies, but when picked and grown in liquid culture they either fail to grow or grow extremely poorly. This happens with multiple vectors for this one construct.
A previous poster asked a similar question, but in the end identified a phage contamination. That is not our problem; only this particular insert is affected. We have tried different cell strains (JM109, TOP10...) and get the same. It is not a problem with the antibiotic, or the vector (other constructs set up at the same time work fine). I cannot see how it can be toxic protein expression, as the vector is for Pichia expression so doesn't contain a bacterial promoter. I am totally stumped as to what is going on.
