I am working with H9C2 cell line and I see difference in morphology of this cells during the culturing. They can be round as well as prolonged ones and it seems to me that the round shape appears when they are stressed.
Sometime after extra passages number as well as extra passage period and other kinds of stress like changes in pH (dropping pH) these cells suffer from some kind of morphological irregularities.
If you find altered H9c2 cell morphology your cells will be under stress, may be due to microbial contamination or due to other reasons. I don't know which passage of cells you are dealing with. any way you can try to recover the cells by increasing the serum in the medium. Before you can go for a live/dead assay.
Indeed the rounded morphology could be a sign of stress due to over crowding, contamination etc. You'd see rounded cells before they actually die. Also over crowding may give you a more elongated myotube morphology which is indicative of differentiation of cells.
As suggested above increase serum levels and adjust cell density. Look out for potential contamination or changes in pH of the medium (color change), temperature of the incubator, water in the incubator etc.
