Im initiating a cell suspension culture from callus of Lupinus mutabilis, the calli I have are friable but once I pass them to a liquid medium they turn brown and compact. I´ve tried adding PVP and celulase but the difference is unnoticable.
Hi Paula, you can play with several factors inorder to reduce the browning. one of such factor is ammonium nitrate ( besides that ammonium is toxic to the invitro growing cells). activated charcol is another potencal alternative to reduce the browning.
Why used liquid medium , you can used semi solid media if you see that liquid media gave you brown and compact callus, Yet, you can addition activated charcoal to liquid media , The hormonal balance in the nutrient medium 'reduces this phenomenon.
