I want to use size exclusion chromatography for drug-encapsulated PLGA nanoparticles. I am going to use Sephadex. but I couldn't find any suitable procedure for nanoparticle separation using SEC. did anyone use SEC for nanoparticles?
In theory yes. In the practice, nanoparticles might interact with the size exclusion medium or become trapped if they can access the largest pores. Moreover, the nature of size exclusion chromatography is the small sample you can process even for a large column. Give it a try and see what happens. If particles are large with respect to the drug I would rather use centrifugation/resuspension or dialysis.
