I have made RPMI 1640 media before 3 days. In tester plate, there are black particles. These particles are big and not like any other contaminant or debries.In higher magnification , they look like FBS particles(thread like structure), but at lower magnification they looks like some floating big dirty black particles ( there is no contamination, no pH change,No turbidity or any other thing due to this). There was 1% PSA in the media. No FBS was added. additionally in our antibiotic stock there were some particles. I have filtered the media again. but still particles are there. They dont affect the cell's growth and morphology.
Please guide me.
Always filter additives to RPMI media to 0.22um or 0.45um, particularly if the particles are big. When using the media, we try to take sterile aliquots of it rather than using it from the bottle - maintains sterility of the stock. While these aren't affecting your cells, I would probably not use the media and try using some reagents from another lab to be sure it won't cause you any troubles.
