Kindly let me know if i can autoclave trypan blue solution as I wish to add it into Agar plates to see clear zones for cellulase activity.
I also have to add CMC to the agar plates, so is 1% CMC solution autoclavable?

I have't used Trypan blue personally. However, I have seen some papers where they use established protocols which mention to use 121 degree autoclaving (on a gentle cycle). However, I have used CMC in my protocols. The polysaccharide has been mentioned to survive at 121 degree autoclaving. However, we generally autoclave this material at 115 degrees for 10 minutes (gentle autoclaving cycle) as we have seen noticeable (although, minor) amounts of hydrolysis on 121 degrees.
Hello. I don't autoclave my trypan blue, as I only used it as a dye. However, I do autoclave CMC.
Autoclave CMC, but in pure water (no divalent cations). CMC solutions are too viscous to be filtered.
