I have't used Trypan blue personally. However, I have seen some papers where they use established protocols which mention to use 121 degree autoclaving (on a gentle cycle). However, I have used CMC in my protocols. The polysaccharide has been mentioned to survive at 121 degree autoclaving. However, we generally autoclave this material at 115 degrees for 10 minutes (gentle autoclaving cycle) as we have seen noticeable (although, minor) amounts of hydrolysis on 121 degrees.
