I am wroking on microbial enzyme and after its extraction in crude form, I had performed the SDS PAGE. But I was not able to get any visible band of protein instead of repeating the whole porocedure several times. What might be the possible reason?

There could be several reasons for it. I wish to know certain more details like, did you see tailing or is that there is no staining at all? What percentage of gel did you use? Additionally, try staining with silver stain.

Yeah Simant Kumar , it is likely that the protein concentration could be one of the reasons. The LOD of silver stain is better and it is more sensitive. Additionally, high salt interferences can also cause the absence of a band.
