Hi. I want to investigate the effect of an herbal drug for memory genes. I selected PC12 cell line. This cell line differentiates neuron-like cells after NGF treatment. Because the NGF is very expensive in my country, I want to select another cell line which be neuronal and don’t need to treat for differentiation. Which cell line do you suggest for my study?

Hi! I am afraid, that any cell line will require some stimulus for differentiation. Occasionaly, you may induce differentiation without expensive growth factors, e.g. when treating a medulloblastoma cell line (SH-SY5Y) with retinoic acid (which is really unexpensive). Apart from that I am not quite sure ,whether a cell line derived from pheochromocytoma (PC12) is the ideal tool to investigate memory processes, but I may be wrong with my opinion. Spontaneously, I would suggest to check, whether primary cell culture techniques may be the better choice for your projects, e.g. chicken neurons, which will differentiate in vitro to a certain degree and are extremely easy to handle with some training (if you are not dependent on cells of human origin).

PC12 cells are a cell line that are used for research of diseases of the brain such as hypoxia, memory dysfunction via prion protein fragments (production of beta amyloid), parkinson disease and addiction studies. you can use from retinoic acid for differetiating. I suggest to you that culture pyramidal cells of rat hippocampus. this primary cell is the best for memory research.

Don't buy it, but if you can get some from your colleagues, try to treat your PC12 with a mix of AMPc + Spermidine (or any polyamine) - check bilbiography for concentrations-. One without the other isn't very effective, but even with the combination, don't expect the same efficiency as when using NGFb.
