For my experiments, where the medium composition is fundamental, I grow all cell lines in the same medium in order to keep comparable basic conditions.
So far my choosen cell lines that have been cultured in a different culture medium from the one that is used for the experiments …..do grow more or less the same.
But what would you suggest, that I do
a) the maintainance in the original medium, let them grow 2 d in the Medium used for the the Experiments, to adapt and then start the exp.?
b) should I also maintain them in the Medium that I use for the experiments…...(which I did so far)
c) when thawing the cells, could I immediatly incubate them in the Medium used for Experiments?!

I don't recommend to change medium for cell lines. For some cells, there is no difference in different medium. But for others, they may be sensitive for what they need. Things might change which can not be just minotored by growth rate. How different are the growth medium and your experimental medium? For each experiment, you always need a negative control which is the one you don't add anything in cells. Why do you have to change medium unless your experiment requires FBS-free or for your convenience?
I mean you need 1 negative control for each cell line. In negative conrol, you use the same medium condition as you are for drug treated cells. The only difference is you don't add testing drug for your negative control. Best to add a positive control too to make sure your drug is working.
Yes I need to change also FBS Levels, it is not for my convenience......
I do always have a negative control, but what I suggest too, is to have a control where you do not add the drug, but only the equal amount of chemical, in which you disolve your drug (i.e. DMSO) ..... so a sample where you have same medium, no drug but only the dissolver.
I guess it is recommended to culture the cells in the same medium in which cells are growing originally. Also it depends on cell lines being used, because few grow well in one medium than others. Hence it is always preferred to grow the cells in the recommended medium. I think you can refer to ATCC website to know the optimal medium for your cells.
