I've a general question as literature is inconsistant: Who has experience with melanoma in vivo models and can help me?
I tried to inject 200 thousand cells s.c w/o Matrigel and started after 8 days, when only black spots but no palbable mass were visible. Still the masses grew so rapidly and partially into the peritoneum that I had to stop after 2 weeks..... I would need to go on for lets say 4 weeks ?!
1) how many cells do you inject where and +/- Matrigel? Coudl it help to keep cells at least initially together ?
2) after how many days do you start your treatment ?
3) how long do you run the experiment?

We do B16 xenografts commercially (http://altogenlabs.com/xenograft-models/melanoma-xenograft/b16-xenograft-model/) and our standard starting dosage of cells in matrigel is 1 million cells into the hind flank. B16 grows pretty quick (you can find growth curves on our website) but other than that we wait for the tumor volume to reach around 100 mm^3 and then begin treatment regimens, which end before the tumors reach 2,000 mm^3. Usually this ends up taking a bit over a month.

We used B16-F10. This model grows very quickly after the tumor grows out (whether use Matrigel or not). By our experience, it usually is less than 2 weeks for the B16-F10 tumors grow from 150mm3 to 2000mm3. It's the characteristics of this model. So to treat for longer time, you have to start treatment very soon after the inoculation.

1- the number of cells depends on your type of your experiment and what you ant to study. And ofc how Lon you want to keep them growing.
Generally speaking, for 200 000 cells you can keep them for over 20 days.
2) Treatment should be started as soon as you observe the tumor. In my many experiments, I have seen people starts between 3 to 5 days.
3) It also depends on the type of experiment and what you study! Plus you want to study which stage of cancer you want to study. I have seen people keep their animals as long as their animal licence allows or when they think keeping more is useless!

@SINA: I want to test a drug and therefore compare tumor growth in treated vs untreated (or Placebo treated mice) .
WHAT do you mean with as soon as one observes ? I first see balckish spots ... but then it rapidly grows.....
Did you also inject less than 200T cells? And do you use matrigel ?

I personally think that the B16 model is less interestig than the B16F10 one (see the attached articles ...).

The problem with the B16 model growing s.c. is the amazing large area of necrosis rendering data difficult to interpret.
