I had already built a xenograft tumor model by employing SK-OV-3 cell line in Balb/c nude mice. And SK-OV-3 worked well. But I didn't test the others. I injected 5X 10^6 cells subcutaneously. The tumor grew up to approximately 5X5MM (length X wideth) about 1~2 weeks. Hope this would help you.
Establishing a xenograft tumor in immune-competent mice (C57bl) would be extremely difficult. You may need to grow these tumors in athymic nude mice as suggested by Kelond Fan.
Yeah, just like Dhatchana Moorthy said, you should use athymic nude mice. If you want use C57bl/6 mice to establish a xenograft model, you can choose a mouse cancer cell line, better the same original to avoid immunological rejection.
Thanks, Dhatchana Moorthy and Kelong Fan! I will do. In my reference was made surgical intervention to inject cell in the ovarian, but if I injected cells subcutaneously I will obtain the same results? Do you have any paper so I can use? thank you.
No, they are different. In your reference, it is a orthotopic transplantation human cancer model of nude mice which means the used human cancer cells are from the same organ you transplanted to the mice. And if you inject the cells subcutanously, it is a xenograft tumor model. You may search the papers used the key words ' orthotopic transplantation' by Google or pubmed. Good luck!
An orthotopic injection of 5x10^5 skov3 cells induce a solid tumor in Nude mice 2 to 3 months post cell injection. If you want to performed a syngenic model in C57B6 mice, you can use id8 cells, orthotopically injected in C57B6 mice which develops tumors under 90 days.
Thanks for suggest, Genevieve Gueguen! I would use ID8 cells in C57B6 mice, but I haven't this cell line available. So I will use Skov-3 in nude, is the best option.
Why is it taking that long 2-3 months? In nu/nu mice tumours should be quickly formed. If you've an issue with tumour formation then you can use Matrigel to be mixed with the cells and then inject it. That will leave the cells in the injection site and tumour growth should be fine in a short timeframe. I hope this helps :).
I should used matrigel in one mice group, but I don't know if Matrigel may be appropriate for this experiment, because a speed growth could increase the oxydative stress and I will measure it.
A PhD student in our lab uses ID8 cells in B6 mice. I can put you in contact with him and he can send you the cell line (just have to agree on the MTA). He is from Belo Horizonte :)
