I want to add PTH(1-34) (Sigma cat# P3796) to some bone cells in vitro.
I cannot find a paper which says how they create a stock solution so I am not sure whether to use ethanol, PBS, DMSO, water, etc for the initial dissolution.
I want my final concentration to be around 1 ng/mL or 250 picomolar. I will do the final dilution of the stock solution in culture media.
Does anyone have experience of what I should initially use?

I used PBS for my experiments, but I went for IP injections instead of adding it to culture media as you intend to do. Worked out for me.
In a more open-minded approach and depending on your time and budget windows, it wouldn't be a bad idea to experiment with the different solvents to check whether there is any difference among them. At the very least, it could be looking at a short communication in a methods journal!

I was PI for Lilly-sponsored, FDA-mandated, 18-month OVX monkey study required for approval of teriparatide (PTH 1-34). Lilly's formulation chemists did tests showing optimum vehicle is physiological saline containing 20 mM NaH2PO4, pH about 4.1. After this monkey study I used this formulation successfully several times for mouse studies. No need for albumin or other components. Stock solution can be sterile filtered.
