I'm going to use PCR method for amplification of a gene from a bacterium. my examples are water and milk That have been infected by a bacterium.
Is there a special kit for the isolation of bacterial DNA from my water and milk samples ? Or before the use the kit should I use other methods such as culture?
I'll guess you can directly extract DNA from the samples and design a couple of primers for each bacterium in order to identify them (there must be a well documented primer database for each strain).

Dear Fahimeh, I agree with Esteban. You can extract DNA either a commercially available kit or a manual method. You should choose appropriate primer set for your target bacteria. To increase number of bacteria, you can enrich your samples in an enrichment broth.

Making a culture will help you get more DNA but you won't be able to grow every strain contained in the oiginal mixture. Some bacteria will outgrow the rest and you won't get any DNA from them.

Dear Fahimeh, there are some commerically available kits to extract DNA from water samples (e.g. from Qiagen). From milk, the following link may help you.

As suggested by earlier comments, you can isolate total DNA from your samples and then design primers to target the bacteria that you suspect to have contaminated the milk and water. This would be an ideal starting point
