Is the MTT result for 72 hour treatment valid if I treat the cells with the drug for 15 days? The cells are exposed to the drugs for far more than 72 hours so how does 72hr MTT treatment result justify that? Help please! thankyou!
The main point is that within 3 hours, 24 hours, and possibly up to 48 hours (though I doubt), the pH of the medium remains stable, and nutrients are abundant. Thus, any observed changes in cellular metabolic activity can be attributed to the test agent. However, after 72 hours (assuming the continous culture), nutrient levels drop and pH decreases. Consequently, it becomes unclear whether changes in metabolic activity are due to the drug or are influenced by environmental stressors.
