How much ug of protein is isolated from 1million of HTR-8/SV Neo cells ?

I am getting 1ug/ul from approx 4million cells, dissolved in 200ul of RIPA, which feels is way lesser. pls help

May somebody help me to standardise the amount and time of the enzyme mixture to isolate Cytotrophoblast from placenta??

i tried with the published protocols, but i am not getting any seperation in the ficol gradient. It seems like i am not incubating the 7gm minced tissue for proper time in the digestion...
