I want to work with brain cell lines which are best to study the effects of different compounds as acetylcholine and butylcholine inhibitors. so far I do not have any experience of working with brain cell lines, please suggest the best cell lines which are easy to study, culture and handle.
Usually SH-SY5Y cells (human neuroblastoma cell line; ATCC number CRL-2266) are used in acetylcholinesterase assays. They are not difficult to handle and culture.

I would second the SH-SY5Y, I've used these cells to study ACh-evoked calcium transients. Beware not to passage them too often or they lose some of their neuronal characteristics...
AChE is not specific cholinergic marker. It is more abundant in postsynaptic noncholinergic neurons or nonneuronal cells with cholinergic innervation. So cholinergic clonal cell line must express ChAT, VAChT and high affinity choline uptake system. They also must accumulate acetycholine and release it in a quantal mode. We used SN (septal neuronal)56 cells (Hammond, Blusztajn). These cells are clearly of cholinergic origin and in native state express ChAT, which may be increased by differentiation with cAMP, retinoic acid, BM9 or several other factors. Other cholinergic cell lines are in native state pluripotential and have much lower levels of cholinergic phenotype than SN56. They must be differentiated to obtain reasonable level of "cholinergecity". Inserts of ChAT gene may yield cells with cholinegic marker levels equal or even higher than in primary brain cholinergic neurons or terminals.
