I am looking for the protocol to evaluate activity of plant extract against Parkinson's disease in in vitro models. If someone have any idea about this, please share with me.
Besides Dr. Neeraj's papers, you also can use PC-12 cells one of the widely used model for PD. Another thing is neuroinflammation also thought to be responsible for PD and AD. So you also can use LPS-induced BV-2 microglial cells to search neuroinflammation.
For plant extracts I recommend a model in vivo: MPTP .... which would inject to the mouse (C57) and within a week the concentration of dopamine is very depressed; this depends on the dose ....
Then you can evaluate the extract..you can measure; apoptosis, oxidative stress, animal viability vs dose, cytokine in serum or be isolated midbrain and make biochemical tests, you can make cuts in the brain and make cytochemical in ventro-medial substantia nigra ...etc etc...
One advice please check de MPTP model in mice C57 bl ( in literature)
