I need to overexpress a microRNA in the livers of normal mice. Can mature miRNA mimics be used for this purpose? If yes, then what additional reagents are required?
Difficult question... Depending on where do you want to have action of your miRNA you can use directly miRNA mimics by tail injection. However, you will expect to have your effect mainly at the liver, and very well irrigated organs. If you want to express your miRNA in other tissues you should think in using viral vectors..
Definetively, if you inject some nucleic acids intravenously in the mouse tail, you will get almost everything within the liver. Of course that you can have some ectopic effects, but for a first approach is the right protocol. This is the principle observed in many small-RNA based drugs like Miravirsen (Santaris Pharma).
The mature miRNA mimics from Dharmacon are excellent choice. However, be sure that you modify the 3' and 5' ends to protect them for degradation. I am sure that Dharmacon people are smart enough to do so.
