I injected MCF7 cells in NOD SCID IL 2 RG null mice, after transplanting an estrogen releasing pellet (not between the shoulders on the back, but on the side). No matrigel, just cells s.c.
The masses grew, even pretty fast in the first 2 weeks after they have been palpable, but now I have the feeling that most of them (90%) grow very slow if not at all and heard about the phenomenon "cachexia". The masses have about 6-7 x 7-8 mm .
What is the cause and how could I avoid it ? Other strain? Maybe the pellet is not working well ?

We've done MCF-7 xenografts commercially (http://altogenlabs.com/xenograft-models/breast-cancer-xenograft/mcf7-xenograft-model/) and you might want to compare your growth curves with ours. We had pretty good growth, but within 12 days they already reach nearly 1000 mm^3. We do use matrigel, and that does help with tumor formation.

I did not really find information about how to diagnose cachexia, but noticed that after a while the majority of the masses start growing again.
Out of 5 mice in my CTR group all developed masses between 3x3mm and 6x7mm roughly so a volume of 60-150mmcubic within 12 days.
Then they kinda stopped for 10-14 days and then 3 grew again, first slow and then much faster, another one started after 6/7 weeks to grow further and the 5th one didn't really grow much .....
Moreover 2 mice (one with the stopped mass, another one with a slow growing one) started to simply loose weight til they where at 60% of the initial weight and I had to sacrifice, but I do not know why ......
