Please recommend a particular catalogue number of 4-VP for protein electrophoresis. We study storage high molecular proteins in wheat and get different results with different 4-vinylpyridine. I will be grateful for comments and discussion.

Eugene, I am working with Pavel. We have been using 2-ME for some time, and the results have been consistent and acceptable. Later, the pattern of the protein bands on the gel changed and it became difficult to read and interpret them. We assume that some conditions/reagents/samples have changed and now disulfide bonds are formed during electrophoresis, when proteins migrate into the gel, and 2-ME or DTT remain in gel loading area. Perhaps adding thioglycolic acid in the upper reservoir buffer can help, but we decided to use a different approach - to modify the free thiol groups of proteins using 4-VP. For some reason, some old 4-VP works, but a fresh reagent does not. There is a stabilizer in 4-VP reagents, and we wonder if some 4-VPs work better than others.
