Since there is no certain way to achieve for plant regeneration systems, trial&error method is only way to check it. However, some combinations of auxin and cytokinins such as BA and NAA, TDZ and IAA (from my research) seem good at for callus and shoot induction, respectively. TDZ is also very effective at low concentrations (0.1 mg/L or less) or its combination with IAA compairing to other auxins promote shoot induction successfully. Similarly, BA is generally not effective (or less efffective) when combined with IAA for shoot induction. BA itself (even at very concentrations up to 10 mg/L), is better a PGR than KIN for callus formation too. In many tissue culture papers, we are seeing synergistic effects of PGR but there is no sufficient causial explanations. So, there seems a big gap in physiological roles of the synergist effects of certain PGR combinations and concentrations in protocols. Of course, there are lots of parameter in culture media, I've just focussed on the effects of auxin and cytokinin together that you know or you would like to offer me(us) for certain species.
