Buhara, we had published the technique for growing Stevia in tissue culture. Eseentially, it is a Murashige& Skoog Medium (MS basal salt medium) spiked with 2% w/v sucrose under two different growth hormones. The first growth hormone tested was 0.1 mg/L of indole-3-acetic acid (IAA). The second growth additive (with natural hormones) was 15%
Thx. Jean. I will try similarmedium composition as you did for germinaion. We tried only MS without any PGR. and germination has not been observed yet. CW is interesting point. ;)
Buhara, based on what we had encountered, your Stevia may be a different variety from ours and many of these varietal differences had not been documented. To obtain good germination, you may need to try out light or dark conditions, lower the MS concentrations to 1/2 or even 1/4 MS, and "trial and error" using a series of gibberellins concentrations. Dear Buhara, Please, use seeds from flowers pollinated by bees during dry growing period, and germinate them at temperature of 25 oC for 10 days, but increase the contact of the seeds with the growing media.
