I am studying the anti cancerous effect of curcumin on cancerous cells (HeLa). I am dissolving the curcumin in DMSO and preparing the subsequent dilutions in DMSO. Is it a correct way or should I use culture media for dilution? To add, irrespective of any working concentration, I am preparing my stock in a such a manner that I am adding only 5 microlitres of curcumin solution for all concentrations. Also, adding 5 microlitres of DMSO, my vehicle control set up, is not causing any deleterious effect on my cell lines.
It s Better to Dissolve High concentration of compound in less volume of DMSO for long storage.
Dilution from stock for cell treatments can be made in medium and store in -20 freezer.
DMSO as vehicle control should not exceed 1% and its better to maintain below 0.1% in total treatment volume.
U can do either way means can prepare subsequent curcumin dilution in DMSO or media but make sure that concentration of DMSO should not be greater than 0.1 %. Even what you are doing is perfectly fine.
